Anti-inflammatory effects of ivy leaves' dry extract

Aim- A study investigated if the anti-inflammatory effect of EA 575® is due to an impact on the NFκB pathway.

Study Design and Method-

  • Immunofluorescence was utilized to observe NFκB nuclear translocation in J774.2 and HEK293 cells.
  • A luciferase-based reporter was used to monitor NFκB transcriptional activity in HEK293 cells.
  • Western blot analysis evaluated the Phosphorylation of RelA and its inhibitor IκB.
  • Protein fragment complementation was used to exhibit changes in the stability of the NFκB:IκB complex. 

Results-

Conclusion: EA 575® can regulate the NFκB pathway, likely by shifting specificity of IKK from IκBα to RelA, thus causing increased stability of NFκB:IκBα complex and decreased RelA translocation into the nucleus.
