Polyethylene glycol 3350 is safe and effective treatment in occasional constipation

The multicenter, double-blind trial evaluated the efficacy and safety of polyethylene glycol (PEG) 3350 in subjects with self-reported occasional constipation.

The study randomized all the eligible subjects ≥ 17 years of age to receive either placebo or PEG 3350 17g once daily and evaluations were done at baseline and after a 7-d treatment period. The primary efficacy variable was the proportion of subjects with complete resolution from straining and hard or lumpy stools, while severity of the daily bowel movement (BM) symptoms and preference of laxatives as per diary entries, visual analog scale scores and questionnaires were the secondary efficacy variables.

Complete resolution was observed in 36.7% of subjects in the PEG 3350 group and 24.5% in the placebo group. Both groups showed similarity in the number of complete BMs without straining or lumpy stools. 

Significant relief in straining and reduction in hardness of stools over a 7-d period was reported in subjects receiving PEG 3350. They also reported a better quality of life with better control over a BM, better relief from constipation, cramping and bloating. PEG 3350 was preferred over other laxatives.

Adverse events (AEs) were similar in PEG 3350 and the placebo groups. 

No deaths or serious AEs were noted and no study participant discontinued treatment due to AEs. 

Thus it was concluded that oral administration of 17 g PEG 3350 once daily for a week is effective, safe, and well-tolerated in subjects with occasional constipation.
