Oxygen radical absorbance of Mulmina Amla

Introduction

Oxygen-free radicals such as the hydroxyl radical, hydrogen peroxide, superoxide anion mediate components of the inflammatory response, with the production of migratory factors, cyclic nucleotides and eicosanoids. An ORAC assay demonstrated the antioxidant activity of Mulmina® Mango. The ORAC assay investigates the free radical damage to a fluorescent probe through the change in its fluorescence intensity.

Objective

To evaluate the total antioxidant capacity of Mulmina® Mango using the ORAC assay.

Methodology

  • The plate was mixed and pre-incubated with a pre-incubation mixture containing sodium phosphate buffer/Trolox/test solution of various concentrations and sodium fluorescein.
  • After pre-incubation, AAPH [2,2’-azobis (2-methylpropionamidine) dihydrochloride] (free radical generator) was added and mixed.
  • The reaction was carried out and the fluorescence measurements were taken at 485 nm excitation and 520 nm emission filters with the following settings:

» Mode: Fluorescence intensity

» Filters: Excitation 485 nm, emission 520 nm

» Plate used: Costar 96, Code 3792 from corning

  • Data reduction and ORAC value calculation were done and the assay was performed in duplicates.

Key observations

  • ORAC value (hydrophilic) for sample Mulmina® Mango was found to be 28.34 µmoles TE/mL
  • The ORAC value of Mulmina® Mango was found to be 5,668/200 mL pack.

Conclusion

Mulmina® Mango contains Mangiferin which is a polyphenol type glycosyl xanthone that exhibits antioxidant properties. Another component in Mulmina® Mango is Centella asiatica that contains triterpene glycosides which are known for their antioxidant effects. The present in vitro study confirmed the antioxidant activity of Mulmina® Mango using ORAC assay.
