The Diabetes Prevention Program (DPP) study results revealed that certain long-term interventions didnt prevent cardiovascular (CV) events among those with prediabetes.

The study enrolled 3,234 individuals with impaired glucose tolerance and divided them randomly into three groups. The first group received 850 mg of metformin twice daily, the second group received a placebo twice daily, and the third group was put under lifestyle modifications. The metformin group continued to receive unmasked metformin, and all patients were offered a less intense group lifestyle intervention.

A follow-up after 18-yr confirmed that metformin and placebo each had comparable effects in reducing the chance of CV events and did not provide cardioprotection effectively (MACE; HR 1.03, 95 % CI 0.78-1.37, P=0.81). Furthermore, participating in a lifestyle intervention did not affect the incidence of MACE compared to placebo (HR 1.14, 95 % CI 0.87-1.50, P=0.34).

Both the measures failed to prevent nonfatal MI, stroke, or CV mortality. The results were reported in the journal Circulation. (MedPage Today May 24, 2022)
